Ebbsfleet United made their numerical advantage count as Cambridge United were unable to fully recover from their early dismissal.
The game started at a frenetic pace with both side's creating a handful of chances to break the deadlock.
However, Cambridge saw themselves reduced to ten men in the 25th minute when Danny Potter received a straight red for a professional foul on Ebbsfleet's John Akinde.
Chris McPhee stepped up to take the resulting spot kick and calmly sent his shot into the bottom-right corner to give his side the lead.
Despite the setback, the visitors drew level seven minutes later when Wayne Hatswell headed a Courtney Pitt free kick in from the centre of the penalty area.
The hosts started to apply the pressure after the break and reclaimed the lead in the 64th minute thanks to a powerful finish from John Akinde.
The U's still find themselves in the promotion play-off zone but they will need to bounce back to winning ways this weekend when they play host to Woking.
