Terry Brown's AFC Wimbledon prepare to take on fellow newcomers Luton Town at on Saturday afternoon for the opening game of the Blue Square Premier campaign.
The Dons boss has doubts over the availability of midfielder Kennedy Adjei, who is struggling with a groin injury.
However, striker Danny Kedwell is available for selection after recovering from a toe injury.
Summer signings Seb Brown, Brett Johnson, Steven Gregory, Derek Duncan and Ricky Wellard are available to make their respective debuts.
Looking ahead to the clash, Brown told the club's official website: "This weekend we look forward to possibly the biggest game in our seven year history.
"It is testimony to everybody at the club and to all the fans that the game is taking place - who'd have thought when we started that we'd be playing Luton in the league in the season 2009-10?
"It gives us the opportunity to flex our muscles against what must be one of the strongest sides in non-league football.
"The next seven days will offer a real insight into what is necessary to compete at this level. Obviously this Saturday's match is one of the most exciting fixtures we could have hoped for.
"Stuart and I have done our homework as far as Luton are concerned and we hope that we can contain what is likely to be one of the most potent strike forces we will encounter this year.
"We shall play our own normal attacking game and whatever happens on Saturday the learning experience for our young squad will be invaluable.
"The players trained last night in what was the fifteenth training session during pre season. During this period we have put a lot of emphasis on fitness and strength.
"It was important to get that in place and we will no doubt get better in other areas, such as set pieces, as the season progresses. I am pleased to report that apart from an injury doubt for Kennedy Adjei we have everybody fit and raring to go."
Luton boss Mick Harford will have to make do without the services of on-loan striker Liam Hatch, after the Peterborough man suffered an ankle injury in training.
However, Steve Basham has subsequently brought in former Exeter striker Steve Basham, who should be available for Saturday's encounter.
Skipper Kevin Nicholls misses out through suspension, along with Alan White, while Ryan Charles is expected to be on the treatment table for the next six weeks after a cartilage operation.
One bit of good news for Harford is that Ed Asafu-Adjaye and Asa Hall are both available after shrugging off respective injuries.
Halford told his club's official website: "I'm really looking forward to it as it seems it's been a long, long time since we started pre-season on 1st July.
"We've played a lot of games and done very well in pre-season. We've worked hard and we look fit and organised.
"On Saturday we'll be up for it and ready for it - backed by a great following of Luton fans.
"It's a challenge that everyone at the Football Club is looking forward to."
